    摘要:

    利用超声法提取黄精总黄丽,选取乙醇体积分数(A)、料液比(B)、超声时间(C)、超声温度(D)进行单因素试验, 在单因素试验基础上,运用 Box-Behnken试验设计,优化提取工艺;应用 DPPH、ABTS及还原力测定等分析各组分抗氧化 能力 。结果表明:黄精总黄丽的最优提取工艺参数:料液比为1:48. 42mL.g - 1,提取时间50. 00min,乙醇浓度53. 33% , 温度54. 66节,在此条件下黄精总黄丽的提取率为30. 12土0. 03mg.g - 1,接近预测值,表明该响应面模型预测性较好;黄 精总黄丽对 DPPH和 ABTS自由基的清除能力在0. 25~3. 00mg. mL- 1 范围内呈递增趋势,具有剂量依赖性,且在质量 浓度为2~3mg. mL- 1 时,清除 ABTS自由基能力与维生素 C效果相当,显示其抗氧化活性较好,可作为良好的天然抗氧 化剂。

    Abstract:

    ultrasonic method was used to extract the total flavonoids of P. sibiricum . Single factor experiments were con- ducted with ethanol volume fraction(A),material liquid ratio(B),ultrasonic time(c),and ultrasonic temperature(D). on the basis of it,Box Behnken Design of experiments was used to optimize the extraction process with the total fla- vonoids extraction rate as the response value;Analyze the antioxidant capacity of each component using DPPH,ABTS, and reducing power measurements . The results showed that the optimal extraction process parameters for total flavonoids in P. sibiricum were as follows:the solid-liquid ratio was1:48. 42mL. g — 1,the extraction time was 50. 00minutes,the ethanol concentration was53. 33%,and the temperature was54. 66节 . under these conditions,the extraction rate of total flavonoids was 30. 12士0. 03mg. g — 1,which was close to the predicted value,indicating that the response surface model had good predictive ability;The scavenging ability of total flavonoids from P. sibiricum to DPPHandABTSfree radicals shows an increasing trend within a certain range,with a dose-dependent effect. At mass concentrations of2mg. mL— 1 and 3mg. mL— 1,the scavenging ability of ABTSfree radicals is equivalent to that of vitamin c. The total flavonoids from P. sibiricum also have good reducing power,indicating good antioxidant activity and can be used as a good natural antioxi- dant.
